Abstract:

Bradykinesia, a hallmark motor symptom of Parkinson’s disease, is characterized by slowness of movement initiation and execution. Its pathophysiology is primarily linked to dopamine depletion in the substantia nigra pars compacta, leading to dysfunction in the cortico-basal ganglia-thalamo-cortical circuits. The reduced dopaminergic input to the striatum disrupts the balance between the direct and indirect pathways, resulting in abnormal oscillatory activity. Here we discuss recent findings on abnormal subcortical and cortical oscillatory activity contributing to bradykinesia. Understanding these mechanisms is crucial for developing targeted therapeutic strategies.
